Description & Mechanism of Action

Tesamorelin is a synthetic analog of growth-hormone-releasing hormone (GHRH), modified with a stabilising N-terminal group to extend its half-life relative to the natural hormone. It binds GHRH receptors on pituitary somatotroph cells with high affinity, stimulating the pulsatile release of endogenous growth hormone (GH) in a pattern consistent with the body's natural secretion rhythm, preserving normal negative-feedback regulation via rising IGF-1 and somatostatin tone. Released GH stimulates hepatic IGF-1 production, which mediates downstream effects including lipolysis — particularly in visceral adipose tissue, which carries a higher density of GH receptors than subcutaneous fat — along with protein anabolism in skeletal muscle and modulation of glucose metabolism. Tesamorelin (brand name Egrifta) holds FDA approval for the treatment of HIV-associated lipodystrophy, and the visceral-fat reduction data referenced in the research literature reflects the clinical trials supporting that approved indication.
