Description & Mechanism of Action
SS-31 (elamipretide) is a synthetic tetrapeptide designed to selectively accumulate in the inner mitochondrial membrane by binding cardiolipin, a phospholipid found almost exclusively in that membrane. By binding cardiolipin, SS-31 is reported to prevent it from converting cytochrome c into a peroxidase, while helping preserve cytochrome c's normal electron-carrying function within the electron transport chain. This interaction has been associated with stabilisation of mitochondrial cristae structure and respiratory chain supercomplexes, reduced mitochondrial reactive oxygen species (ROS) production, and improved ATP synthesis and membrane potential in stressed cells, including cardiomyocytes and neurons in preclinical models. Because its mechanism centres on membrane electrostatics and cardiolipin binding rather than a classical receptor interaction, SS-31 is studied primarily as a tool for probing mitochondrial bioenergetics and oxidative stress rather than a conventional receptor-mediated pathway.
