Description & Mechanism of Action
Ipamorelin is a synthetic pentapeptide and a selective agonist of the ghrelin receptor (GHS-R1a), structurally distinct from GHRH analogs such as tesamorelin. It binds GHS-R1a on pituitary somatotroph cells, triggering a Gq/11-mediated phospholipase C signalling cascade that raises intracellular calcium and drives exocytosis of stored growth hormone (GH). Its defining research characteristic is selectivity: in the landmark 1998 characterisation study, ipamorelin stimulated GH release without producing significant elevations in cortisol, prolactin, ACTH, FSH, LH or TSH — a selectivity profile that distinguished it from earlier, less selective growth hormone secretagogues such as GHRP-6. Because it acts through the ghrelin receptor rather than the GHRH receptor, ipamorelin is frequently studied alongside GHRH analogs (e.g. CJC-1295, tesamorelin) for their complementary, non-overlapping stimulation of the GH/IGF-1 axis.
