Description & Mechanism of Action
Epithalon (Epitalon) is a synthetic tetrapeptide (Ala-Glu-Asp-Gly), modeled on a naturally occurring peptide bioregulator of the pineal gland, developed through Russian peptide-bioregulator research beginning in the 1980s. Two distinct mechanisms have been proposed in the literature: first, induction of the telomerase catalytic subunit (TERT) in cultured human cells, reported to extend telomere length and, by extension, cellular replicative capacity; second, a pineal-signalling effect in which Epithalon upregulates phosphorylated CREB (pCREB), increasing expression of AANAT, the rate-limiting enzyme in melatonin biosynthesis, and thereby modulating circadian gene expression. A landmark rodent lifespan study reported a 12.3% increase in both mean and maximum lifespan in Epithalon-treated mice compared to controls. The evidence base for Epithalon is predominantly Russian preclinical and rodent research, with limited independent international human-trial validation.
Applications in Research
- Studying telomerase (TERT) induction and telomere-length maintenance
- Pineal gland and melatonin-synthesis (AANAT/pCREB pathway) research
- Rodent lifespan and aging-biomarker research models
- Circadian gene expression studies
Handling, Reconstitution & Stability
- Handle as a hygroscopic powder; open vials and weigh under dry conditions where possible
- Reconstitute with sterile water or bacteriostatic water, adding solvent slowly down the side of the vial
- Swirl gently to dissolve — avoid vigorous shaking
- Filter-sterilise (e.g. 0.22 µm) immediately before use if required by your protocol
- Aliquot reconstituted solution and store at -20°C or below; avoid repeated freeze-thaw cycles
Precautions & Notes
- Evidence base is predominantly Russian preclinical and rodent research, dating to the 1980s-90s, with limited independent international human-trial validation
- Experimental effects are concentration-, duration-, and model-dependent
- Include appropriate experimental controls to distinguish specific from non-specific effects
- For laboratory research use only — not for human or veterinary use
