Description & Mechanism of Action

CJC-1295 (No DAC), also known as Modified GRF 1-29, is a 29-amino-acid analog of growth-hormone-releasing hormone (GHRH) engineered with amino acid substitutions for improved stability against enzymatic degradation, but without the Drug Affinity Complex (DAC) modification that allows the related long-acting variant to bind serum albumin and remain active for days. Without DAC, this version acts through the same GHRH-receptor pathway on pituitary somatotrophs — stimulating pulsatile growth hormone release — but with a much shorter half-life, requiring more frequent dosing in research protocols than the DAC-modified form. Because it shares its receptor pathway with other GHRH analogs such as tesamorelin, it is frequently studied as a reference compound for GHRH-receptor pharmacology, and is often compared alongside ghrelin-receptor agonists such as ipamorelin, which stimulate GH release through a separate, complementary pathway.
