Description & Mechanism of Action
5-Amino-1MQ is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that consumes nicotinamide and S-adenosylmethionine (SAM) to produce 1-methylnicotinamide (1-MNA) and S-adenosylhomocysteine (SAH). By inhibiting NNMT, 5-Amino-1MQ is reported to reduce this methylation "sink," increasing SAM availability for other cellular methylation reactions while freeing nicotinamide for recycling toward NAD+ via the NAMPT salvage pathway. The resulting increase in NAD+ availability has been studied for its support of redox and sirtuin-dependent pathways linked to mitochondrial function, lipid oxidation, and insulin action. In diet-induced obesity mouse models, selective NNMT inhibitors including 5-Amino-1MQ have been reported to reduce white adipose tissue mass and body weight and improve several metabolic parameters.
