Description & Mechanism of Action

5-Amino-1MQ is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that consumes nicotinamide and S-adenosylmethionine (SAM) to produce 1-methylnicotinamide (1-MNA) and S-adenosylhomocysteine (SAH). By inhibiting NNMT, 5-Amino-1MQ is reported to reduce this methylation "sink," increasing SAM availability for other cellular methylation reactions while freeing nicotinamide for recycling toward NAD+ via the NAMPT salvage pathway. The resulting increase in NAD+ availability has been studied for its support of redox and sirtuin-dependent pathways linked to mitochondrial function, lipid oxidation, and insulin action. In diet-induced obesity mouse models, selective NNMT inhibitors including 5-Amino-1MQ have been reported to reduce white adipose tissue mass and body weight and improve several metabolic parameters.

Applications in Research

  • Studying NNMT enzyme inhibition and methyl-group metabolism
  • NAD+ salvage pathway and sirtuin-linked research models
  • Diet-induced obesity and adipose-tissue research
  • Investigating SAM availability and cellular methylation capacity

Handling, Reconstitution & Stability

  • Supplied as a solution — no reconstitution required
  • Store as directed on the product label, protected from light
  • Use aseptic technique when drawing from the vial to avoid contamination

Precautions & Notes

  • Experimental effects are concentration-, duration-, and model-dependent
  • Include appropriate experimental controls to distinguish specific from non-specific effects
  • For laboratory research use only — not for human or veterinary use

This product is supplied for in vitro research purposes only. It is not intended for human or animal administration, diagnostic use, or any other purpose. Handle in accordance with good laboratory practice guidelines.
